Cover: Lysosomal permeability after incubation with glycated collagen as assessed by decreased co-localization of dextran-Texas red with Lamp-1. Human umbilical vein endothelial cells (HUVEC) were incubated for 24 h in culture medium containing dextran conjugated to Texas red (MW 10,000) followed by a 24-h chase in a dextran-free culture medium for accumulation in lysosomes. Cells were incubated with glycated collagen (GC), native/glycated collagen (1:3 GC), native collagen (NC), or remained untreated (control) and studied at the indicated times. Displayed is a cell treated with chloroquine (300 mM), which was used as a positive control. Cells were fixed, permeabilized, and stained with antibody to Lamp-1 and a nuclear stain, Hoechst 33342. Cells were imaged using a Nikon TE-2000U microscope equipped with a Spot Insight camera (Diagnostic Instruments).
